GPR150, also known as PGR11, is an orphan class A GPCR belonging to the vasopressin/oxytocin receptor-related subfamily. The receptor is very poorly characterized pharmacologically, and no endogenous ligand or validated G-protein coupling pathway has yet been established. Current evidence does not identify any confirmed agonist, antagonist or endogenous ligand for GPR150. GPR150 expression appears to be relatively restricted, although its tissue distribution in humans has not yet been comprehensively characterized. The receptor has attracted some attention in cancer biology, as altered GPR150 expression and promoter methylation have been associated with ovarian cancer. Beyond these observations, the physiological function of GPR150 remains largely unknown, with no clearly established role in neuronal, endocrine or metabolic regulation. No selective small-molecule agonists or antagonists have been convincingly described, and there are currently no established pharmacological tool compounds for this receptor. Consequently, no approved drug or clinical-stage compound targeting GPR150 is currently available. Overall, GPR150 represents a highly unexplored orphan GPCR with considerable potential for future ligand discovery and target validation, particularly in oncology, but substantially more research is required to define its pharmacology and physiological function.
