Threonine311/Serine312 is a major phosphorylation site of the A3 Adenosine receptor. The pT311/pS312-A3 antibody detects phosphorylation in response to high-efficacy agonists but not after PKC activation. T311/S312 phosphorylation is a key regulator of A3 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the A3 Adenosine Receptor (A3) and high level of GRK2 were either not exposed or exposed to 100 nM NECA for 30 minutes. Cells were lysed and immunoblotted according to 7TM Western Blot Protocol with the anti-pT311/pS312-A3 antibody (7TM0021A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-A3 antibody to confirm equal loading of the gel.
